L-phenylalanine applications and culture duration affect root growth and production of tropane alkaloids and phenolics in adventitious root cultures of Hyoscyamus niger L.

This study was aimed to determine the effects of different concentrations L-phenylalanine (0.25, 0.50 and 1.00 mM L-Phe) applied adventitious root cultures for culture durations on growth production tropane alkaloids phenolics in Hyoscyamus niger. For this purpose, roots obtained from petiole explants vitro seedlings were used as plant materials. After cultured liquid Murashige Skoog (MS) medium, L-Phe roots. Then harvested 1st, 3rd, 7th day applications, fresh dry weights, indexes analysed. It also examined contents (hyoscyamine scopolamine) (gallic acid, catechin, epicatechin, vanillin, cinnamic rosmarinic p-coumaric, o-coumaric rutin, caffeic ferulic chlorogenic quercetin) by HPLC both medium. As a result, according parameters data, day*application interaction not found statistically significant. The maximum scopolamine hyoscyamine amounts detected with application 3 days. In terms phenolics, 1st 3rd days selected most appropriate applications duration. Consequently, it has been determined that secondary metabolite may be increased H. niger selection durations. is an effective inexpensive metabolites correct concentration
